Development of an ELISA for sensitive and specific detection of IgA autoantibodies against BP180 in pemphigoid diseases.

In pemphigoids, a group of autoimmune skin blistering diseases collagen XVII/BP180 is a major target of autoantibodies (aAbs) belonging to the IgG, IgE and IgA isotypes. For the detectionn of BP 180-specific circulating IgG and IgE aAbs several ELISAs were developed. In the present study we used a recombinant form of BP 180 ectodomain to establish the first sensitive and specific ELISA for the detection of BP 180-specific IgA aAbs from sera of patients with the lamina lucida-type of linear IgA disease (n= 30) and healthy donors (n=105). The test´s specificity was 83.3%. This new ELISA should prove a useful tool for clinical and translational research as well as for the diagnosis and disease monitoring of patients with IfA pemphigiod.
