How mammalian cells decode continuous TGF-? ligand doses into discontinuous cellular fate decisions remain poorly understood. Using a combined experimental and mathematical modeling approach, we found that the TGF-? pathway displays different sensitivities to ligand doses at different time scales: short TGF-? signaling responses are graded, but long-term TGF-? signaling responses are switch-like to a small change in TGF-? levels, which might be critical for cell fate determination.
