Kidins220/ARMS associates with B-Raf and the TCR, promoting sustained Erk signaling in T cells

The Ras?MAPK pathway is crucial in decision processes that cells take upon stimulation. In T cells one member of this pathway, B?Raf, is activated upon stimulation of the T cell antigen receptor, leading to sustained MAPK signaling and T?cell effector functions. Using mass spectrometry, we identified protein kinase D?interacting substrate of 220 kDa (Kidins220) as a novel B?Raf interaction partner, and found that it is also associated with the pre?TCR, ??TCR and ??TCR. We showed that Kidins220 is dispensable for TCR?induced transient?, but is required for sustained Erk activation, for calcium signaling, CD69 upregulation, IL?2 and IFN?? production. In conclusion, Kidins200 is a novel and crucial signalling molecule in the immune system.
