In this BIOSS review, we summarize the current knowledge on the activation of the T-cell antigen receptor (TCR) by the natural antigens: foreign peptides presented by major histocompatibility complex molecules (MHCp) and bacterial superantigens. MHCp that binds with sufficient affinity to TCR?? thereby elicits phosphorylation of the cytoplasmic tails of the TCR?? -associated CD3 subunits. This is the initial step in T cell activation.
