Deciphering the epigenetic code of cardiac myocyte transcription.

Epigenetic processes are essential for cell identity and gene regulation. To identify mechanisms of transcriptional control, cell type-specific analysis is required. The aim of our study was to purify cardiac myocyte nuclei from frozen hearts and to determine RNA expression as well as epigenetic profiles. RNA sequencing from nuclei revealed a higher degree of unspliced RNA and a higher abundance of genes involved in transcriptional regulation as compared to results obtained from cardiac myocytes. An integrative analysis of nuclear mRNA and histone mark occurrence indicated a major impact of chromatin state on gene expression in cardiac myocytes. This study establishes cardiac myocyte-specific sorting of nuclei and our data provide novel insight into cardiac myocyte transcription.
