We show that two phylogenetically distant viruses, a double stranded DNA virus, herpes simplex virus-1 (HSV-1) and a single stranded RNA virus, Semliki Forest virus (SFV) use the same BH3-only protein Puma to mediate Bax/Bak activation, mitochondrial outer membrane permeabilization and apoptosis in human and mouse cells. Puma is regulated on the posttranscriptional and transcriptional level, the first occurring rapidly after viral infection, the second probably acting as apoptosis amplification.
