AKT-dependent phosphorylation of the SAM domain induces oligomerization and activation of the scaffold protein CNK1

Scaffold proteins such as CNK1 are hubs for the coordination of intracellular signaling networks. Here, we demonstrate that AKT-dependent phosphorylation of the sterile motif alpha (SAM) domain induces oligomerization of CNK1 in growth factor-stimulated cells and subsequently CNK1-mediated signaling. Searching the COSMIC database revealed the AKT phosphorylation site as putative target for oncogenic activation of CNK1 leading to constitutive CNK1 oligomerization and CNK1 signaling.
