Here, we describe the development of the first green light-responsive system for the targeted control of gene expression in mammalian and plant cells. We validate functionality in a variety of mammalian and plants cells and provide a model-based, quantitative characterization of the system’s performance. Using green light as stimulus is of special interest in plant backgrounds as endogenous plant photoreceptors have minimal activity in this part of the light spectrum.
