Lipase Congeners Designed by Genetic Code Engineering

Applying a ‘genetic code engineering’ approach we substituted all methionine, proline, phenylalanine and tyrosine residues with related synthetic analogs in Thermoanaerobacter thermohydrosulfuricus lipase. The generated lipase congeners were endowed with chemical diversity not found in nature and displayed special features such as enhanced activation, elevated enzyme activity, substrate tolerance, as well as changes in optimal temperature and pH.
