In this BIOSS publication we reviewed old and new technical developments in studying signal transduction mechanisms in immune cells using synthetic biology approaches. These include rebuilding of small functional signaling systems and the design of new molecules acting as signaling switches. In addition we describe how sophisticated engineered immune receptors (chimeric antigen receptors) are being tested in the clinic for gene therapy against tumours.
