Dynamic gene expression is often controlled by multiple cis-regulatory modules (CRMs) but how the activities of CRMs are coordinated in time and space is yet unclear. With Drosophila follicle cell patterning as a model and a combination of genetics and computer modeling we show that an early CRM of the gene broad initiates a cascade of events that protect a late CRM from premature EGFR-mediated repression. The work provides essential insights into dynamics of gene expression and CRM-landscape organization.
