The cellular microenvironment is shaped by soluble factors and structural components, the ECM. A quantitative mass spectrometry strategy was developed to assess proteome differences in the microenvironment of human fibroblasts. The influence of collagen VII (C7), a structural component, which is lost in genetic skin fragility, was used as model. Loss of C7 had a global impact on the microenvironment and was associated with proteome alterations highly relevant for disease pathogenesis.
