We use genome editing approaches and quantitative imaging to understand gene regulation with Drosophila as a model system. We focus on brinker, a key developmental gene, which is transcriptionally repressed by TGFb-signalling through multiple silencer elements (SE) to generate an instructive protein gradient. Our work provides insights in enhancer organization and a solid experimental framework for future studies in this direction.
