Control of E-cadherin adhesion is essential for gastrulation. Song et al. show that Pou5f1/Oct4 mutant zebrafish embryos accumulate E-cad at the plasma membrane. Mutants lack EGF expression, which in turn controls proper E-cad endocytic trafficking. The dynamics of E-cad trafficking and adhesion is essential for effective cell migration during gastrulation.
