Cellular stress induces Bax-regulated nuclear bubble budding and rupture followed by nuclear protein release

Here we show a novel regulatory mechanism of apoptosis, the Bax-mediated generation and subsequent rupture of nuclear protein-containing bubbles in response to cellular stress. The bubbles do not contain DNA and are encapsulated by impaired nuclear pore-depleted nuclear envelopes. The process precedes apoptosis and occurs independently of caspases. Surprisingly, the activity of Bax is non-canonical as it depends on the BH3-domain and alpha helices 5/6 of Bax, but it is not inhibited by Bcl-xL.
